Fenbendazole
Fenben, or Panacur as it is also known, is an antihelmintic of the benzimidazole type. Since its discovery in 1975, it has been used worldwide. In the past few decades, fenbendazole and similar drugs have shown a significant impact on cancer. (Ref.1, Ref.2)
This led to the theory that this de-wormer listed for dogs could be used as an anti-cancer medicine. In fact, the benzimidazole-class drugs can perform in a similar way to a wide range of contemporary chemotherapy medications such as Taxol and Vinca alkaloids. ( Refer. ) Available toxicological data suggests that fenbendazole is well tolerated by humans after oral contact. However, there are only a few human studies. (Ref.)
Fenbendazole powder is usually available unadulterated and has a white color with a weak solubility in water.
This drug is not well absorbed in the bowels. Food in the stomach may increase bioavailability.
What is the mechanism of action of fenbendazole?
Fenbendazole is a drug that was originally intended to treat parasites. It works by inhibiting microtubule production through a specific attachment to b-tubulin. This prevents the polymerization and death of parasite cells by b-tubulin dimers.
It's amazing to see that fenbendazole and other benzimidazoles have a similar effect on tumor cells. There are currently three main methods that fenbendazole uses to destroy cancer cells.
Induction of apoptosis. The anti-tumor effects are thought to be achieved by combining the medicine with b-tubulin. This leads to the stopping of the cell cycle and is cytotoxic.
It has been shown that benzimidazole-carbamates can stop tubulin polymerization in microtubules in helminths and human tumor cells. (Ref.1, Ref.2)
The use of glucose by cancer cells is known to be excessive. The Warburg effect means that cancer cells use glucose 200 times faster than normal cells. PET scans reveal this – metabolically active sites that consume additional radioactive glucose are clearly visible and believed to be cancerous tumors or inflammation.
Fenbendazole reduces sugar consumption by cancer cells through a reduction in GLUT transporters, which are channels that deliver glucose from the blood into cancerous cancerous cell. It also inhibits the enzyme Hexokinase 2. Hexokinase is essential for cancer cell survival. It helps tumours grow by producing more sugar and accelerating lactic acidosis in the extracellular matrix. In addition, restarting p53 prevents the production of fuel for cancer. When p53 returns, it stops the expression and action of GLUT1 (glucose gate) transporters in cells. (Ref.)

There is a growing amount of research that has confirmed that fenbendazole can actually boost the strongest tumour suppressor in the body – the p53. (Ref.)
Elephants have twenty copies of the gene p53 in their genome, but humans only have one. It's strange that an elephant gets cancer less frequently than a person. These animals are large, but they also have a greater number of cells and a higher amount of genetic mutations.
It is interesting to note that cancer cells are able to adapt to the de-wormer drug and escape it. This occurs in a number of chemotherapy and biological therapies.
Cancer cells can become resistant to chemotherapy when they adapt to the ability to excrete medications by themselves, through drug efflux pumps called P-glycoproteins. Fenbendazole doesn't target p-glycoproteins and therefore can't be ejected from cancer cells once it's in there.
The tumors will not develop any resistance to fenbendazole. It is still effective and will continue to destroy cancer cells. This is not the case with paclitaxel or other chemotherapies such as vinblastine, Docetaxel, Vincristine, etc. (Ref.)

Side effects and safety of Fenbendazole
